Botryosphaeria
dieback, esca, and eutypa dieback are three economic major grapevine trunk diseases causing severe yield reduction in vineyards worldwide (Bertsch et al., 2013). The frequency of disease symptoms has increased considerably over the past few decades and currently, no efficient treatment is available to control these diseases. Botryosphaeria dieback, namely Black Dead Arm (BDA), is associated with the development of fungi from the Botryosphariaceae family in the wood of diseased grapevines. Vitis vinifera cultivars show different levels of susceptibility to grapevine trunk diseases, generally established by the extent of foliar symptoms apparition in the field. However, the cause of these foliar symptoms and the relation between foliar symptoms development and wood necrosis remain only partially understood.


We have investigated the resistance level by monitoring necrosis symptoms in the Vitaceae family after artificial inoculation of Neofusicoccum parvum and Diplodia seriata on woody internodes. Three species from V. vinifera subsp. Sylvestris, the ancestor of V. vinifera, were selected based on a lower necrosis development after inoculation with N. parvum and D. seriata. Expression of defense genes after inoculation was studied. To know if V. vinifera is able to perceive the metabolites synthesized by fungi associated to BDA, an actin marker line of grapevine V. vinifera Chardonnay GFP-AtFABD2 10a was used, since actin filaments has been reported as a sensor or in processing/decoding in early plant innate immunity (Guan et al., preparing). Christophe Schmidt (March-Sep., 2015) and Hugo Ralay Rakotonarivo (March-Augus, 2016) from UHA France have been conducted their master practical course in SW Uni China under the frame of the metabolite profiling of Vitis suspension cell lines after infection by fungi of the Botryosphaeriaceae family, which will accumulate evidences for better understanding the plant-pathogen interaction during the infection of Botryospheriaeace.
